What Polynucleotides Are

PDRN — polydeoxyribonucleotide — is short, purified DNA fragments, historically derived
from salmon milt because its sequences are highly compatible with human DNA. Newer
microbial-derived sources are emerging. The purified material contains no cells, no proteins,
and nothing capable of transferring genetic material.

Its mechanism is well characterised: PDRN acts on the adenosine A2A receptor and feeds the
cellular salvage pathway, producing anti-inflammatory signalling and support for tissue
repair. It also forms a hydrating gel-like scaffold that supports fibroblast activity.

The distinction that matters clinically: this is a repair and regeneration
treatment, not a volumising or hydrating one.

The Under-Eye Indication — Why This Matters

This is where polynucleotides earn their reputation, and where they solve a genuine problem
for a large group of patients.

Thin, crepey under-eye skin is extremely common, and hyaluronic acid filler is frequently
the wrong answer for it — HA binds water, so in this tissue it risks persistent puffiness, and
placed superficially under thin skin it can produce a blue-grey Tyndall discolouration. Anyone

Polynucleotides work differently. Low-viscosity formulations are designed to thicken and
repair periorbital skin rather than fill a hollow — improving crepey texture and skin quality
without adding volume, without water-binding bulk, and without the Tyndall risk. Published
work on periorbital lines is among the strongest evidence in this field.

So the routing is straightforward: a genuine hollow needs filler; thin,
crepey, poor-quality under-eye skin needs this. Many patients who were told they were not
candidates for tear trough filler are good candidates for polynucleotides.

Fish Allergy — Read Before Booking

Traditional polynucleotide products are salmon-derived. Purification removes proteins, but
a fish or seafood allergy must be declared and assessed by the doctor before any treatment is
planned. Do not leave this until you are in the chair.

Risks

Common and expected: injection bumps for up to 48 hours, redness, bruising,
mild swelling and tenderness.

Less common: persistent small nodules; longer-lasting bumps in very thin
periorbital skin; infection at an injection point; and rarely an inflammatory reaction.

Important difference from HA: polynucleotides are not hyaluronic acid, so
hyaluronidase does not dissolve them. There is no reversal agent. They do
resorb naturally, and the volumes involved are very small, so the practical consequences are
limited — but it is worth knowing that the undo option available with HA filler does not
apply here.

Rare: vascular complications. Risk is low given superficial placement and
tiny volumes, but not zero, and this should be performed by a medical injector.

Who Should Not Have It

Not suitable if you have a fish or seafood allergy that has not been
medically assessed and cleared; are pregnant or breastfeeding; have an active skin infection,
inflamed acne or cold sore in the area; or have a history of severe allergic reactions or
